Recent research indicates a potential breakthrough treatment for advanced breast cancer. Studies focus on trastuzumab rezetecan, a drug targeting ERBB2-low metastatic breast cancer, which is characterized by low levels of HER2, a protein influencing cancer cell growth.
Clinical Trial Insights
The phase 3 trial, HORIZON-Breast06, evaluated the drug’s effectiveness against standard chemotherapy in this cancer type. Conducted across 73 hospitals in China, involving 551 female patients, the trial ran from June 2023 to August 2025. Participants received either the drug intravenously every three weeks or standard chemotherapy.
The results were promising. Patients using trastuzumab rezetecan experienced a median progression-free period of 16.4 months, compared to 6.8 months with chemotherapy. Additionally, their likelihood of cancer progression or death during the study was reduced by 73%.
Research Findings
Published in JAMA, the study showed tumor reduction in 75.5% of patients receiving the drug, versus 41.2% with chemotherapy. At 12 months, 90.6% of patients on trastuzumab rezetecan were alive, as opposed to 81.4% on chemotherapy.
Trastuzumab rezetecan provides significant progression-free survival benefit over standard chemotherapy for ERBB2-low advanced breast cancer.
Dr. Loren Rourke, a breast surgical oncologist, described the findings as “incredibly encouraging” for patients, offering more time without disease progression. However, she highlighted the importance of weighing side effects, such as anemia and blood-related issues, against the benefits.
Side Effects and Future Research
Serious side effects occurred in over 84% of patients treated with the drug, compared to nearly 68% with chemotherapy. While side effects were “generally manageable,” more research is required to understand them, especially those affecting blood counts and increasing infection risks.
Dr. Rourke emphasized the need to explore whether survival duration is extended by the new treatment. Although it delays cancer progression, it remains vital to assess its impact on overall lifespan.
These findings illustrate a significant step forward in breast cancer treatment, with ongoing research required to answer remaining questions.
