New Treatment Shows Promise for Endometrial Cancer

Merck’s latest investigational drug, sacituzumab tirumotecan (sac-TMT), has displayed promising results in treating endometrial cancer during a Phase 3 clinical trial. The company’s announcement highlighted the drug’s success in achieving its primary endpoints of overall survival and progression-free survival in patients with advanced or recurrent endometrial cancer.
The TroFuse-005 trial marks a significant milestone, being the first global Phase 3 trial to demonstrate statistically significant improvements in survival compared to chemotherapy in these patients. Notably, sac-TMT is the first antibody-drug conjugate (ADC) to achieve this result in endometrial cancer treatment.
An ADC is a specialized cancer therapy that targets tumor cells more effectively, minimizing harm to healthy cells. Sac-TMT is administered bi-weekly through intravenous infusion.
The trial involved 776 patients whose condition worsened after undergoing both platinum chemotherapy and immunotherapy. These patients were randomly assigned to receive either sac-TMT or a physician’s choice of treatment, including doxorubicin or paclitaxel chemotherapy.
Results indicated that those treated with sac-TMT showed clinically meaningful improvements compared to other treatment options. The study also met response rate benchmarks and presented similar side effects as seen in previous trials of the drug.
Although specific statistics regarding survival benefits, response rates, and side effects were not disclosed, the data is expected to be presented at an upcoming medical conference. Dr. Domenica Lorusso, the study’s lead investigator, stated that sac-TMT may address a critical unmet need for certain patients with advanced endometrial cancer.
Dr. Brian Slomovitz, co-director of gynecologic oncology at Mount Sinai, highlighted the rising number of endometrial cancer cases and related deaths, surpassing those of ovarian cancer in the United States. Despite advancements in immunotherapy, better treatment options for recurrent disease remain an unmet need.
If future data confirm the initial findings, the key considerations will focus on the extent of survival benefits and the drug’s toxicity profile, which will determine sac-TMT’s place in the treatment landscape. An overall survival improvement offers real benefits to patients and their families.
